Quantitative FCS in Zero-Mode Waveguides at Micromolar Concentrations

New article and Application Note explore ZMW-FCS measurements and programmable workflows with Luminosa

Comparison of fluorescence detection in a diffraction-limited confocal volume and a zero-mode waveguide (ZMW) nanoaperture. At elevated fluorophore concentrations, the conventional confocal observation volume contains a large number of molecules (N > 1000), whereas confinement of the excitation field within the ZMW reduces the effective observation volume and consequently the molecular occupancy (N < 10), enabling FCS measurements at higher concentrations.

Zero-mode waveguides (ZMWs) reduce the effective observation volume, enabling FCS measurements at micromolar fluorophore concentrations while maintaining low molecular occupancy.

Our latest article shows how Luminosa combines ZMW imaging and positioning with quantitative FCS. It also explores programmable multi-ZMW measurements with the LumiPy Python interface, demonstrated in a proof-of-concept experiment across 88 ZMWs.
